When the Kalki 2898 AD film was announced in August 2019, Prabhas was revealed as the film's lead actor by the production house on 26 February 2020. In July 2020, actress Deepika Padukone (her Telugu debut) was signed to play the lead role opposite Prabhas.

After Deepika, Amitabh Bachchan joined the cast in a full-length role in the film in October 2020. Disha Patani joined the cast in May 2022, Kamal Haasan in June 2023, and Anna Ben joined the cast (her Telugu debut) in February 2024.
